EVOO Polyphenols Exert Anti-Inflammatory Effects on the Microglia Cell through TREM2 Signaling Pathway

چکیده
In Alzheimer’s disease (AD), microglia, brain resident immune cells, become chronically inflammatory and neurotoxic. recent years, neuroinflammation has attracted particular interest in the scientific community. The genetic variants of molecules associated with ‘‘microgliopathies’’, including triggering receptor expressed myeloid cells-2 (TREM2), result increased risk developing AD cognitive decline. We performed a set vitro assays using human neuronal (SH-SY5Y) microglial (BV2 C13NJ) cell models. Cells were differentially treated extra virgin olive oil (EVOO) polyphenols, oleuropein aglycone (OleA) hydroxytyrosol (HT) before adding LPS. evaluated protective effects these EVOO products by biochemical biology assays, ELISA, MTT, ROS detection, Western blotting immunofluorescence. Our results provide an integrated understanding neuroprotection exerted polyphenols terms of: (i) reduction pro-inflammatory cytokines release (IL-6, IL-8, IP-10 RANTES); (ii) activation TREM2-dependent anti-inflammatory pathway; (iii) enhancement activity favoring M2 polarization phenotype. Such findings new important insights into mechanisms which dietary exert beneficial properties against impairment.

TREM2/DAP12 Complex Regulates Inflammatory Responses in Microglia via the JNK Signaling Pathway
DNAX-activating protein of 12 kDa (DAP12) is a signaling adapter protein expressed in cells that participate in innate immune responses. By pairing with different triggering receptors expressed on myeloid cell (TREM) proteins, DAP12 can mediate both positive and negative cellular responses. In particular, TREM1 acts as an amplifier of the immune response, while TREM2 functions as a negative reg...

Glycyrrhiza glabra is a plant used in traditional medicine across the world for its pharmacological value. Its value has been proved such as anti-cough, anti-diabetes, anti-microbial and antioxidant effects. The roots of plant comrised glycyrrhizin which is 50 times sweeter than sugar. Macrophages can stimulated during inflammatory disorders, and with production of multiple inflammatory mediato...
متن کاملAnti-Inflammatory Effects of α-Galactosylceramide Analogs in Activated Microglia: Involvement of the p38 MAPK Signaling Pathway
Microglial activation plays a pivotal role in the development and progression of neurodegenerative diseases. Thus, anti-inflammatory agents that control microglial activation can serve as potential therapeutic agents for neurodegenerative diseases. Here, we designed and synthesized α-galactosylceramide (α-GalCer) analogs to exert anti-inflammatory effects in activated microglia. We performed bi...
متن کاملAnti-inflammatory Signaling in Microglia Exacerbates Alzheimer’s Disease-Related Pathology
In this issue of Neuron, Chakrabarty et al. (2015) and Guillot-Sestier et al. (2015) reveal that the anti-inflammatory cytokine IL-10 inhibits Aβ clearance by microglia, worsening cognitive decline in mouse models of Alzheimer's disease (AD). These studies provide further support that pro-inflammatory signaling is an innate immune defense mechanism in AD.
متن کاملGlobular Adiponectin Limits Microglia Pro-Inflammatory Phenotype through an AdipoR1/NF-κB Signaling Pathway
We recently reported that increased levels of Adiponectin (ApN) in the brain led to microglia phenotype and activation state regulation, thus reducing both global brain inflammation and depressive-like behaviors in mice. Apart from this, little is known on ApN molecular effects on microglia, although these cells are crucial in both physiological and pathological processes. Here we fill this gap...
